Indications for Use:
Tramadol is an opioid analgesic indicated for the management of pain that has not responded to standard therapies. These may include both pharmacological interventions, such as NSAIDs and paracetamol, as well as non-pharmacological approaches. It is often used in cases of postoperative pain, chronic pain conditions, and injury-related discomfort.
Mechanism of Action:
Structurally related to codeine and morphine, tramadol acts through dual mechanisms:
Opioid Receptor Binding: Tramadol weakly binds to ยต-opioid receptors in the brain, increasing the threshold for pain perception and enhancing pain tolerance.
Monoamine Reuptake Inhibition: It also inhibits the reuptake of noradrenaline and serotonin in the central nervous system. This action disrupts the transmission of nociceptive (pain) signals at the level of the spinal cord, providing an additional analgesic effect.
This dual mode of action makes tramadol effective in managing complex and treatment-resistant pain.
